Endocrine potential of genistein, resveratrol and 8-prenylnaringenin in gonadal and thyroid axes and related organs

Three phytoestrogens belonging to different structural classes - the isoflavonoid genistein, the stilbene resveratrol and the flavanone 8-prenylnaringenin - were investigated for their endocrine disrupting potential. The substances were tested for their effects on LH and TSH release in pituitary cell culture, and for the response of the HPG- and HPT-axes in ovariectomized rats after a single dose challenge or after long term treatment for three months. Changes in related organs like the reproductive tract, metabolic activity of the liver and bone condition were investigated, too. All three compounds displayed significant and varying changes in the different test systems. Caution is advisable regarding the long term consumption of these naturally occuring compounds as nutritional supplements or herbal preparations at high concentrations without thorough risk-benefit assessment.
