Charakterisierung der Wechselwirkungen zwischen dem Notch-Signalweg und intrinsischen und extrinsischen Signalen in hämatopoetischen Zellen

Der Notch-Signalweg ist ein wichtiger Regulator von Differenzierungsprozessen, Proliferationsvorgängen und der Apoptose. Die zugrundeliegenden molekularen Mechanismen, die eine so breite Wirkungspalette ermöglichen, sind jedoch weitgehend unverstanden. Ziel dieser Arbeit war es, Notch1 und dessen Zusammenspiel mit verschiedenen Interaktionspartnern in hämatopoetischen Zellen genauer zu charakterisieren. Unter Verwendung der multipotenten hämatopoetischen Zelllinie FDCPmix, die ein Tamoxifen-induzierbares System der konstitutiv aktiven intrazellulären Domäne des Notch1-Rezeptors trägt, sollte der Einfluss des Notch1-Rezeptors auf die Differenzierung und Proliferation hämatopoetischer Zellen untersucht werden. Im ersten Projekt wurde die Wirkung von Notch1 auf die Erythropoese analysiert. Notch1 beschleunigte dabei die erythroide Differenzierung und erhöhte die Expression von β-Globin, indem der Notch1-Signalweg über das Effektormolekül RBP-J die hypersensitive Stelle 2 (HS2) der Locus-Kontroll-Region des β-Globins aktiviert. Die Expression der erythroiden Transkriptionsfaktoren Gata1, Fog1, Eklf und Nfe2 wurde dagegen durch die Aktivierung von Notch1 nicht beeinflusst. Das zweite Projekt beschäftigte sich mit der funktionellen Interaktion des Notch1-Signalweges mit p53, einem wichtigen Faktor der Tumorsuppression und des Proliferationsarrests. Die Aktivierung von Notch1 in hämatopoetischen Zellen führte zu einem p53-abhängigen Zellzyklusblock. Dabei wurde durch Notch1 die Expression des Zellzyklusregulators p21Waf/Cip in p53Wildtyp, nicht aber in p53null FDCP-mix-Zellen erhöht. Die Regulation der p21Waf/Cip-Expression durch Notch1 erfolgte hierbei nicht über RBP-J, sondern über verstärkte Bindung von p53 an den p21Waf/Cip-Promoter. Eine Wiedereinbringung des p53-Gens in p53null Zellen stellte den Notch1-vermittelten Zellzyklusblock wieder her. In einem weiteren Projekt wurde der Einfluss von Notch1 auf Caspasen, die für apoptotische Prozesse verantwortlich sind, untersucht. Die in Vorarbeiten gezeigte Regulation von Caspase-6 mRNA durch Notch1 konnte in dieser Arbeit auf Proteinebene weder für Caspase-6 noch für Caspase-3 bestätigt werden. Jedoch wurde die Expression von Caspase-7 durch Notch1 verstärkt, was allerdings nicht zu einer erhöhten Apoptoserate führte. Weitere Forschung muss hier aufzeigen, ob Notch1 über Caspasen eine Wirkung auf Differenzierungsprozesse hat und wie diese molekular vermittelt wird.

Notch1 is an important mediator of cell fate determination, proliferation and apoptosis. However, the underlying molecular mechanisms facilitating this broad range of impact are largely unknown. The aim of this work was the characterization of the Notch1 signaling pathway and the molecular interactions between Notch1 and its interaction partners in hematopoietic progenitor cells in detail. Multipotent FDCP-mix cell lines carrying a conditional tamoxifen-inducible system of the constitutively active intracellular domain of Notch1 were used to analyze the effects of activated Notch1 on differentiation and proliferation processes in hematopoietic cells. In the first part, the effect of Notch1 on erythropoiesis was examined. Notch1 was shown to accelerate erythroid differentiation and upregulate β-globin expression by activating the hypersensitive site 2 (HS2) of the β-globin locus control region via the Notch1 effector RBP-J. In contrast, the expression of the erythroid transcription factors Gata1, Fog1, Eklf and Nfe2 was not altered after activation of Notch1. The issue of the second project was the interplay of Notch1 with p53, a critical factor of tumor suppression and growth arrest. It was demonstrated that activation of Notch1 led to a p53-dependent inhibition of the cell cycle in hematopoietic cells. Furthermore, Notch1 upregulated the cell cycle inhibitor p21Waf/Cip in p53wild-type, but not in p53null FDCP-mix cells. Induction of the p21Waf/Cip gene by activated Notch1 was mediated by a RBP-J independent pathway, but involved increased binding of p53 to the p21Waf/Cip promoter. The re-expression of p53 in p53null cells restored the Notch1-mediated inhibition of proliferation. In another project the effect of Notch1 on caspases, a group of proteins responsible for apoptotic processes, was analyzed. Previously, Notch1 was shown to activate caspase-6 mRNA expression. The upregulation of both caspase-6 and caspase-3 by Notch1 could not be confirmed at the protein level. In contrast, caspase-7 was found to be activated by Notch1, though this activation did not result in an enhanced apoptosis. Further work is needed to analyze if Notch1 regulates differentiation processes via activation of caspases and how this is mediated.
