Die Adenosin-Rezeptoren als Target zur Synthesemono- und bizyklisch annelierterPyrimidine

Die Entwicklung potenter sowie selektiver Adenosin-Rezeptor-Antagonisten hat in den letzten Jahren immer mehr an Bedeutung gewonnen. Einige daraus hervorgegangene Substanzen befinden sich bereits in klinischen Studien zur Überprüfung ihrer Effizienz bei der Behandlung von u. a. Herzinsuffizienz und Morbus Parkinson. Unter den in vorangegangenen Arbeiten dargestellten Pyrido[2,3-d]pyrimidinen und Pyrimido[2,1-f]purindionen finden sich hochpotente A1-Rezeptor-Antagonisten. Zur Aufstellung neuer Struktur-Wirkungs-Beziehungen an den Adenosin-Rezeptoren wurden im Rahmen der vorliegenden Arbeit unter Einbezug der für die Substanzklassen der Pyrido[2,3-d]pyrimidine und Pyrimido[2,1-f]purindione entworfenen pharmakophoren Modelle zahlreiche neue Synthesewege zur Darstellung 1,3-Dimethyl-1,2,3,4-tetrahydropyrimidin-annelierter, bi- und trizyklischer Heterozyklen entwickelt. Dadurch konnte eine Vielzahl von Substanzen mit den folgenden unterschiedlichen Zielstrukturen realisiert werden: Furo[2',3':4,5]pyrido[2,3-d]pyrimidine, Imidazo[5',4':4,5]pyrido[2,3-d]pyrimidine, Imidazo[5,4-e]pyrimido[2,3-b]diazepine, enantiomerenreine Pyrimido[4,5-b]azepine, Pyrazolo[3',4':4,5]pyrido[2,3-d]pyrimidine und Pyrrolo[2',3':4,5]pyrido[2,3-d]pyrimidine. Für die dargestellten Verbindungen erfolgte eine umfassende Strukturaufklärung insbesondere unter Verwendung ein- und zweidimensionaler NMR-Spektroskopie-Methoden. Zur Bestimmung der Enantiomerenreinheit der Pyrimido[4,5-b]azepine wurde eine HPLC-Methode entwickelt. In Kooperation mit der Arbeitsgruppe von Prof. Dr. C. E. Müller des pharmazeutischen Instituts der Universität Bonn wurde eine Auswahl der dargestellten Verbindungen an den Adenosin-Rezeptor-Subtypen A1 und A2A in Radioligand- Bindungsstudien getestet. Dabei stellte sich das 5,7-Dibenzyl-1,2,3,4,5,6-hexahydro- 6H-imidazo[5',4':4,5]pyrido[2,3-d]pyrimidin-2,4,6-trion als ein potenter und im Vergleich zu den Pyrimido[2,1-f]purindionen selektiver A1-Rezeptor-Antagonist heraus. Folglich wurde eine neue Leitstruktur für die Entwicklung einer neuen Substanzklasse potentiell hochwirksamer A1-Rezeptor-Antagonisten gefunden.

Adenosine receptors play an important role in the pathogenesis of many diseases such as heart failure and Parkinson’s disease. In the last decade extensive research efforts have been directed towards the development of highly effective and selective modulators of the adenosine receptor family. Some of these compounds are currently in clinical trials. The development of potent A1 receptor antagonists like pyrido[2,3-d]pyrimidines and pyrimido[2,1-f]purindiones has led to pharmacophore models for these core moieties. The goal of the thesis was to use the information obtained from these two models to synthesize 1,3-dimethyl-1,2,3,4-tetrahydropyrimidine annelated bicyclic and tricyclic heterocyclic compounds in order to explore the structure-activity relationships on adenosine receptors, especially the A1 receptor. This effort has led to the following compound classes: furo[2',3':4,5]pyrido[2,3-d]pyrimidines, imidazo[5',4':4,5]pyrido[2,3-d]pyrimidines, imidazo[5,4-e]pyrimido[2,3-b]diazepines, enantiopure pyrimido[4,5-b]azepines, pyrazolo[3',4':4,5]pyrido[2,3-d]pyrimidines and pyrrolo[2',3':4,5]pyrido[2,3-d]pyrimidines. All the synthesized compounds were analyzed by comprehensive and detailed structure elucidation methods, particularly by the means of one- and twodimensional NMR-spectroscopy. In addition, a high-perfomance liquid chromatography method was developed to determine the enantiopurity of pyrimido[4,5-b]azepines. In collaboration with Prof. Dr. C. E. Müller’s group at the University of Bonn assorted compounds were tested in radioligand binding assays for A1 and A2A adenosine receptor inhibition. 5,7-Dibenzyl-1,2,3,4,5,6-hexahydro-6H-imidazo[5',4':4,5]pyrido[2,3-d]pyrimidin-2,4,6-trion was found to be a very potent and selective A1 receptor antagonist, even more selective than the pyrimido[2,1-f]purindiones. This newly synthesized lead compound could be exploited by decorating with appropriate substituents to design highly potent and selective adenosine receptor antagonists.
