Kohlenhydrat-Pinzetten:Synthese photoaktivierbarer Pseudoamidglycoside für die H-Brücken-unterstützte Photomarkierung

Die Aufklärung mechanistischer Details molekularer Erkennungsprozesse auf zellulärer Ebene und insbesondere die Identifikation der hierfür entscheidenden Bindedomänen stellt eine große Herausforderung dar. Die Photoaffinitätsmarkierung kann hierzu einen nützlichen Beitrag leisten. Eine Steigerung der Ausbeute im entscheidenden Quervernetzungsschritt der Photomarkierung ist von großem Interesse, um die Isolation und Identifikation kovalent verknüpfter Ligand-Protein-Fragmente nach enzymatischer Hydrolyse zu erleichtern. Daher wurde im Rahmen der vorliegenden Dissertation ein Kohlenhydrat-basiertes System entwickelt, das durch gezielten Einbau von Harnstoff- bzw. Thioharnstoff-Teilstrukturen über Wasserstoff-Brücken-Bindungen eine Präorganisation erlaubt bzw. komplementäre Strukturen binden kann. In Kombination mit photoaktivierbaren Gruppen (z. B. Diazirinen) und integriert in Liganden-Strukturen könnte dieses System zur Effizienzsteigerung von Photomarkierungsexperimenten in biologischen Zusammenhängen führen. Gezielt funktionalisierte Trifluormethylaryl-diazirine zur Thioharnstoff- bzw. Harnstoff-Kupplung wurden synthetisiert und ihre photochemischen Charakteristika betrachtet. Darüber hinaus wurde ein neuartiger, perfluorierter Alkydiazirin-Baustein als Photophor dargestellt, der in einen Kohlenhydrat-Liganden eingebaut werden kann. Es konnten diverse 1,6-Bis-Thioharnstoff-funktionalisierte Kohlenhydrat-Pinzetten synthetisiert werden. An diesen Verbindungen wurden 1H-NMR-spektroskopische Untersuchungen zur Beurteilung ihrer Wasserstoff-Brücken-Bindungseigenschaften in verschiedenen Lösungsmitteln und in Mischung mit Glycin-Harnstoff-funktionalisierten Gast-Verbindungen durchgeführt. Es konnte basierend auf den Ergebnissen gezeigt werden, dass die Möglichkeit besteht, die Bindung eines Bis-Thioharnstoff-modifizierten Liganden an ein Rezeptor-Protein anhand der chemischen Verschiebung der NH-Protonen im NMR zu verfolgen. Eine Fluor-Substitution sowie 15N-Isotopenmarkierung in den Thioharnstoff-Teilstrukturen ermöglicht in diesem Zusammenhang die eindeutige Zuordnung der detektierten Thioharnstoff-NH-Signale auch in Lösung neben Proteinen. Des Weiteren wurde der Einfluss des Substitutionsmusters auf die E/Z-Isomerenaufspaltung innerhalb der 1,6-Thioharnstoff-Kohlenhydrat-Pinzetten untersucht und ergab für Aryl-Substitution an den Thoharnstoff-Teilstrukturen eine für die konzeptionierte Komplexierung bzw. Präorganisation günstige Beeinflussung der Thioharnstoff-Strukturen untereinander. Weiterhin konnten zwei unterschiedliche photoaktivierbare 1,6-Bis-Thioharnstoff-funktionalisierte Kohlenhydrat-Pinzetten synthetisiert werden. Zusätzlich wurden Syntheserouten etabliert, die zu C1- und C6- bzw. nur C6-Position elongierten, photosensitiven Kohlenhydrat-Konjugaten mit Bis-Thioharnstoff- und alternativ Bis-Harnstoff-Teilstrukturen führen. Zusätzlich zu den Kohlenhydrat-Pinzetten wurden verschiedene N-basierte Pinzetten-Systeme als Verknüpfungseinheit synthetisiert. Es wurden neben den photochemischen Charakterisitika bei Diazirin-Funktionalisierung der bidentalen Bis-Thioharnstoff-Dendrite auch die Komplexierungs-Eigenschaften Aryl-substituierter Dendrite mit aromatischen Glycin-Harnstoff-Gästen in einem 1H-NMR-Mischungsexperiment untersucht.

The elucidation of mechanistic details of molecular recognition processes at the cellular level and especially the identification of the fundamental recognition domain represents a major challenge. The method of photoaffinity labeling can therefore make a big contribution. An increase in the yield at the critical cross-linking step of photolabeling is of great interest to facilitate the isolation and identification of covalently linked ligand-protein fragments after enzymatic hydrolysis. Therefore, in the context of this thesis a carbohydrate-based system was developed, which by selective incorporation of urea or thiourea moieties allows preorganization or can bind complementary structures via hydrogen-bridge bonds. In combination with photoactivatable groups (e.g. diazirines) and integrated into a ligand structure such a system could be used to increase the efficiency of photolabeling experiments in biological contexts. Several Trifluormethylaryldiazirines specifically functionalized for thiourea or urea-coupling were synthesized and their photochemical characteristics were studied in detail. In addition, a novel perfluorinated alkyl diazirine building block was prepared, which can be installed in a carbohydrate ligand structure. Various 1,6-bis-thiourea functionalized carbohydrate tweezers were synthesized. 1H-NMR spectroscopic studies were conducted of these compounds to assess their hydrogen-bonding properties in different solvents and in mixtures with glycine-urea-functionalized host structures. Based on the results it could be shown, that it is possible to follow the binding of a bis-thiourea-modified ligand to a receptor protein on the basis of the chemical shift of NH protons in the NMR. A fluorine-substitution and 15N-isotope labeling in the thiourea-substructure would allow the unambiguous assignment of the detected thiourea NH signals next to proteins in solution. Furthermore, the influence of the substitution pattern on the E/Z-isomer distribution within the 1,6-thiourea-carbohydrate tweezers was investigated and found that aryl substituents at the thiourea-substructures shows a reciprocal interaction of the thiourea structures at C6- and C1-position and thus could advantage the conceptualized complexation or rather preorganization. Two different photoactivatable 1,6-bis-thiourea functionalized carbohydrate tweezers were synthesized. Furthermore synthetic routes are established, which lead to C1- and C6- and only C6-position elongated photo-sensitive carbohydrate conjugates with bis-thiourea- and alternatively bis-urea-substructures. Apart from the carbohydrate molecular tweezers various N-based systems have been synthesized as a linking unit. Next to the photochemical characteristics in case of diazirine-functionalization of the bis-bidentate thiourea dendrite, the complexation properties of a aryl-substituted aromatic dendrite with glycine-urea guests were examined.
