Makrocyclen durch dynamisch-kombinatorische KondensationsreaktionenImine - Hydrazone - Oxime

Im Rahmen dieser Arbeit sollten Makrocyclen durch dynamisch-kombinatorische Konden-sationsreaktionen erzeugt werden. Daher wurden 15 verschiedene divalente Bausteine synthetisiert, die zu 15 dynamisch-kombinatorischen Hydrazon- bzw. Oxim-Bibliotheken (DCLs) kombiniert wurden. Für diese Bausteine wurde zum einen das 2,6-disubstituierte Pyridin-Grundgerüst und zum anderen das Oligoethylenglycol-Grundgerüst gewählt. Das 2,6-disubstituierte Pyridin wurde Aldehyd-, Hydrazid- und Hydroxylamin-funktionalisiert. Die Oligoethylenglycole wurden zu Dihydrazinen, Dihydraziden und Dihydroxylaminen, mit jeweils drei verschiedenen Kettenlängen, umgesetzt. Zusätzlich wurden ein Dibenzaldehyd-Baustein und zwei Alkan-Dihydroxylamine synthetisiert. Jeweils ein Pyridin-Baustein wurde in Kondensationsreaktionen mit den unterschiedlich langen komplementären Bausteinen zu Hydrazonen und Oximen umgesetzt. Aufgrund der divalenten Struktur der Bausteine konnten sich in den DCLs makrocyclische und lineare oligomere Spezies bilden. Die Bausteine besitzen durch ihre Stickstoff- und Sauerstoffatome jeweils mehrere Donor-Atome, die Metallionen komplexieren können. Die mit den synthetisierten Bausteinen aufgebauten dynamisch-kombinatorischen Bibliotheken wurden auf die Amplifizierung von Makrocyclen durch verschiedene Metallionen als Template untersucht, wofür NMR, MS und HPLC-MS-Analytik genutzt wurden. Als Ergebnis wurde festgestellt, dass die [1+1]-Makrocyclen ausgehend von den Pyridindicarbaldehyden und den Oligoethylenglycol-Ketten sowohl mit als auch ohne Templationen bevorzugt gebildet wurden. Mit den jeweiligen längeren Oligoethylenglycolderivaten bildeten sich die Makrocyclen generell besser. In einigen Fällen wurden Makrocyclen durch die schwereren Erdalkalimetallionen amplifiziert. Darüber hinaus wurden drei verschieden lange Pyrrol-basierte Diamine synthetisiert, die in Kondensationsreaktionen mit dem Pyridindialdehyd zu Iminen umgesetzt wurden. Dadurch bildeten sich ditope Imin-Makrocyclen, die auf ihre Fähigkeit zur Ionenpaar-Erkennung untersucht wurden.

In this thesis, macrocycles should be formed by dynamic combinatorial condensation reactions. Therefore, 15 different divalent building blocks were synthesized, whereby 15 dynamic combinatorial libraries were generated. As building blocks, two scaffolds were chosen: the 2,6-disubstituted pyridine and oligo¬ethylene glycols. The pyridine scaffold was synthesized with aldehyde, hydrazine and hydroxylamine functio¬nalities. The oligoethylene glycols were converted to dihydrazines, dihydrazides and dihydroxylamines, each of them with three different chain lengths. Furthermore, a dibenzaldehyde and two alkane-dihydroxylamines were synthesized. One pyridine building block and one of the chain building blocks were used in condensation reactions to form hydrazones and oximes. As a consequence of the divalent structures, the formation of both macrocyclic and linear oligomeric species was possible in the DCLs. Due to the aldehyde functionality, which can be on either of the building blocks, different orientations of the C=N-double bond were obtained. The nitrogen and oxygen atoms in the building blocks serve as donor atoms which can be used for the complexation of metal ions. The composition of the DCLs was investigated in the presence of different template metal ions. NMR, MS, and HPLC-MS were employed as analytical tools. The pyridine aldehyde and the NH2-functionalized oligoethylene glycols formed [1+1]-macrocycles as favoured products without templates as well as with templates. It was observed that the macrocycles of the longer oligoethylene glycols were formed more favourably. In some cases macrocycles were amplified by the heavier alkaline earth metal ions. Furthermore, three diamines with various lengths based on a pyrrole-scaffold were synthesized, reacting with a pyridine aldehyde to imines in condensation reactions. The resulting ditopic imines may bind cations and anions and were therefore examined for their ability of molecular recognition of ion pairs.
