Proteinexpressionsanalyse und biochemische Funktionsanalyse von Hornerin und Trichohyalin-like1 in der Haut des Menschen

Trichohyalin-like1 und Hornerin gehören zu den SFTPs, die in Haut und den Anhangsorganen exprimiert sind. Diese Proteine bestehen aus einer N-terminalen S100-Domäne, einem Spacer und einer unterschiedlichen Anzahl von Sequenzwiederholungen. TCHHL1 konnte im distalen Teil der inneren Haarwurzelscheide detektiert werden. Der Nachweis des natürlichen Proteins in Extrakten war nicht möglich. Ursache könnte eine veränderte Antigenität oder eine erschwerte Extraktion durch die enzymatische Umwandlung der Arginine in Citrulline oder die enzymatische Ausbildung von Isopeptidbindungen sein. HRNR-Fragmente zeigen eine hohe Flexibilität, sowie die Abwesenheit typischer Sekundärstrukturelemente. HRNR wurde durch bioinformatischen Berechnungen, Ergebnissen aus der CD-Spektroskopie, seiner hohen proteolytischen Empfindlichkeit und weiterer Eigenschaften der Gruppe der „intrinsically disordered proteins, IDPs“ zugeordnet. Für verschiedene HRNR-Fragmente konnte antimikrobielle Aktivität gegen E. coli, P. aeruginosa, S. aureus und C. albicans nachgewiesen werden. Die hydrophoben AS haben keinen und die Arginine haben nur gegen E. coli einen Einfluss auf die antimikrobielle Aktivität. Teilfragmente zeigten vergleichbare antimikrobielle Aktivitäten. Die Salzkonzentration hatte den größten Einfluss auf die antimikrobielle Aktivität. Beim hautphysiologischen pH-Wert von 5,5 und Nährstofflimitierung zeigte sich eine deutliche Steigerungen der antimikrobiellen Aktivität. In elektronenmikroskopischen Untersuchungen wurden elektronendichte Aggregate nach Behandlung mit HRNR detektiert. Die Ergebnisse lassen vermuten, dass HRNR eine wichtige Komponente des Haut-Abwehrsystems darstellt, das durch Generierung hochaktiver Proteinfragmente die gesunde Haut vor Infektionen schützt. Die artifizielle Induktion des HRNR und die Nutzung von HRNR-Fragmenten als Antiinfektiva könnten daher zukünftige Optionen für die Behandlung infektiöser Hauterkrankungen sein.

Trichohyalin-like1 and Hornerin belongs to the SFTP-family, which are expressed in skin and skin-appendages. These proteins consist of an N-terminal S100-domain, a spacer, a different number of repeats and a C-terminal part. TCHHL1 could be detected in the distal part of the inner root sheath. The immunological detection of the natural protein isolated from whole skin, epidermis, callus or isolated hair follicles extracts by western blot analyses failed. The enzymatic conversion of arginine to citrulline by PAD 1 or the enzymatic formation of isopeptide bonds between glutamine and lysine by TGase 1 results in an associated change in the antigenicity and a hindered extraction due to cross-linking in vitro HRNR fragments show high flexibility as well as the absence of typical secondary structure elements. HRNR was assigned by bioinformatics, results of CD spectroscopy, its high proteolytic sensitivity and other characteristics to the group of "intrinsically disordered protein, IDPs". For various HRNR fragments antimicrobial activity against E. coli, P. aeruginosa, S. aureus and C. albicans could be detected. The hydrophobic AA didn’t affect the antimicrobial activity, arginine only influence the activity against E. coli and smaller fragments showed similar antimicrobial activity compared to the longer ones. The concentration of salt shows the highest impact on the antimicrobial activity. The skin physiological pH of 5.5 and a nutrient limitation, increase the antimicrobial activity. Electron dense aggregates were detected by electron microscopy after treatment with HRNR. The results suggest that HRNR represents an important component of the skin-defense system, generating highly active protein fragments, which protect healthy skin by infection. The artificial induction of HRNR and the use of HRNR fragments as anti-invectives could therefore be future options for the treatment of infectious skin diseases.
