Toll-Like Receptor Expression Profile Of Gingival Mesenchymal Stem Cells In Inflamed And Non-Inflamed Conditions

BACKGROUND: Gingival margin-derived stem/progenitor cells (G-MSCs) display notable periodontal regenerative potential in vivo. During the regenerative process, G-MSCs may interact with their inflammatory microenvironment through toll-like-receptors (TLRs). The current study intended to depict the expression profile of G-MSCs’ TLRs. MATERIAL AND METHODS: Cells were isolated from free gingival margins, STRO-1-immunomagnetically sorted and then seeded to attain single colony forming units (CFUs). G-MSCs were characterized for the markers CD14, CD34, CD45, CD73, CD90, CD105, CD146 and the expression of STRO-1, besides the multilineage differentiation potential. Following G-MSCs' incubation in basic (non-inflammatory) or inflammatory medium (IL-1β, IFN-α, IFN-γ, TNF-α) a TLR expression profile was created. RESULTS: G-MSCs displayed all characteristics of stem/progenitor cells. In basic (non-inflammatory) medium G-MSCs expressed TLRs 1, 2, 3, 4, 5, 6, 7, and 10. The inflammatory milieu significantly up-regulated TLRs 1, 2, 4, 5, 7 and 10, but diminished TLR 6 (p≤0.05, Wilcoxon-Signed-Ranks-Test). CONCLUSIONS: This study pronounces for the first time the distinctive TLRs expression profile of G-MSCs in inflamed and non-inflamed milieus.
