In this thesis it was demonstrated that the mitochondrial amidoxime reducing component is able to reduce hydroxamic acids. The reaction is catalyzed by the protein partners cytochrome b5 reductase and cytochrome b5. Hydroxamic acids display an important pharmacophore in a variety of drugs (e.g. antibacterial, cytostatic, antiinflammatory), so their metabolism is of great interest for the development of new and existing drugs. Via the hydroxamic acid function, multivalent metal cations can easily be chelated. The reduced form of hydroxamic acids, the amide, is not able to do this. Thus, the reduction of hydroxamic acids by mARC leads to inactivation of the drug substance. The following protein sources were used for the investigation: subcellular fractions, recombinant proteins, cell culture experiments and organ homogenates of mARC2 knockout mice. Therefore, reduction by mARC should always be considered in the development of new drugs with hydroxamic acid function.
