Planung und Aufbau von Flugzeit Massenspektrometern und Verwendung der Massenspektrometrie zur Konformationsanalyse

This work reports the construction of two time-of-flight mass spectrometers. As a basis for construction the theory of ToF-MS is developed first. Consequently the planning, simulation and first spectra of a miniaturized ToF-MS with new reflector design are described. The "Mini-ToF" works in REMPI and LEI mode with resolution power of ca. 200. The second instrument that was built is a MALDI-ToF-MS which is a slightly modified model of an existing instrument. The reflector was optimized. As an application of mass spectrometry the possibilities to analyse conformational states of peptides and proteins are reviewed. Two methods of conformational analysis with mass spectrometry are applied to beta-amyloid. The results do not indicate the expected conformational change. Possible reasons are discussed.
