Untersuchung von klinischen, biochemischen, histologischen und immunhistochemischen Markern des Therapieansprechens in Patienten mit chronisch entzündlichen Darmerkrankungen unter IL-6 trans-signaling Blockade (Olamkicept) : Retrospektive Analyse aus einer Phase IIa Studie

IL-6 trans-signaling is known as a proinflammatory signaling pathway in inflammatory bowel diseases (IBD). Olamkicept, a selective IL-6 trans-signaling inhibitor, was given to patients with IBD for the first time as a part of a phase IIa study.

Compatibility and effects of Olamkicept on the inflammatory activity were assessed in a multimodal way with special focus on immunhistochemical changes in the gut.

In a subgroup Olamkicept induced clinical and biochemical remission as well as mucosal healing. Immunhistochemically, antiinflammatory effects occured, especially concerning proliferation and presence of immune cells. Mucosal STAT3 phosphorylation, being the target of Olamkicept, was blocked significantely. pSTAT3 is considered as an early and specific marker for therapeutic response on Olamkicept.
In a subgroup of IBD, IL-6 trans-signaling seems to be an increased signaling pathway and this subgroup may profit of the treatment with Olamkicept in particular.
