Miniaturization of Proteomics Sample Preparation by Digital Microfluidics

Approaches for mass spectrometry-based proteomics have experienced significant sensitivity improvements in recent years, making the analysis of limited sample amounts feasible. However, due to factors such as manual pipetting errors and the large volumes of reactants used, conventional proteomics protocols are not applicable for low-input proteomic sample processing. In nanoproteomics, sample preparation methods are tailored to the analysis of minute amounts of sample or even single cells. In this work, digital microfluidics as a liquid handling technology was introduced, adapted, and optimized for miniaturized proteomics sample preparation. This comprised four experimental sub-projects focusing on the development of suitable sample preparation steps and technical adaptations for digital microfluidics. The primary objective was to achieve both qualitative and quantitative proteome analyses of very small sample quantities.
