Das Laserablationsverhalten von ionischen Flüssigkeiten verschiedener MALDI-Matrices

Gegenstand der vorliegenden Arbeit ist die Untersuchung des Ablationsprozesses von ionischen Flüssigkeiten als Matrix in der UV-MALDI. Die Verwendung dieser sogenannten ionischen flüssigen Matrices ist eine recht neue Entwicklung und zeigt in einigen Bereichen Verbesserungen gegenüber konventionellen kristallinen Matrices. Da einige Vorteile der Flüssigmatrices auf einen gleichmäßigeren Ablationsvorgang zurückgeführt wurden, ist die Untersuchung der dabei zugrundeliegenden Mechanismen von großem Interesse für die zukünftige Entwicklung weiterer Verbesserungen dieser Methode. Die erste Methode, die zur Charakterisierung der Ablation verwendet wurde, ist die Postionisation von neutralen Molekülen in der Ablationswolke. Diese stellen den Großteil des ausgestoßenen Materials dar und die Bestimmung der mit dieser Methode zugänglichen Parameter, die Geschwindigkeitsverteilung und die Translationstemperatur der Teilchen, ermöglicht eine quantitative Beschreibung des Ablationsprozesses. Hier konnte gezeigt werden, dass der Ablationsvorgang der verwendeten Matrices sehr gut reproduzierbar ist und diese Stoffe sich daher gut für die Untersuchung mit dieser Methode eignen. Weiterhin konnte festgestellt werden, dass für ionische Flüssigkeiten der häufig verwendeten Matrices 2,5-DHB und CHCA sehr ähnliche Ablationsparameter erhalten werden, die deutlich unter den Werten für die jeweiligen festen Stoffe liegen. Zusätzlich konnten Einflüsse der Lichtabsorption bei der verwendeten Ablationswellenlänge und der Energie der eingestrahlten Photonen gefunden werden. In den Experimenten zeigte sich auch das Auftreten eines zweiten Ablationsvorganges neutraler Moleküle um einige 100 ns verzögert zur initialen Ablation. Die Charakterisierung dieses Prozesses und die Aufklärung seines Ursprungs war mit der Postionisationsmethode nicht möglich. Um dies zu erreichen, wurde ein neues Experiment entworfen und in Betrieb genommen. Dieses kombiniert ein Flugzeitmassenspektrometer mit der Fast Flash Imaging Methode, bei der Schattenbilder der ablatierten Teilchenwolke zu verschiedenen Zeitpunkten nach dem Ablationslaserpuls mit Hilfe sehr kurzer Lichtpulse im Nanosekundenbereich aufgenommen werden. Anhand der hiermit durchgeführten Messungen konnte ein Modell entworfen werden, welches das Auftreten der verzögerten Ablation durch die verspätete Freisetzung von einem Teil der Energie, die durch den Laserpuls in die Probe eingebracht wurde, erklärt und die auftretenden Änderungen im Ablationsvorgang bei Variation der laser fluence und der Ablationswellenlänge zufriedenstellend erklären kann.

The subject of this work is the investigation of the ablation process of ionic liquids used as the matrix in the UV-MALDI. The utilization of these so called ionic liquid matrices is a recent developement and has been shown to exhibit advantages in some areas of work in comparison to the conventional crystalline matrices. Because some of the advantages of the liquid matrices have been explained by an increase of the uniformity of the ablation event, the investigation of the underlying mechanisms is of great interest for prospective developments of further enhancements of the method itself. The first method that has been used to characterize the ablation is the postionization of neutral molecules in the ablation plume. These neutrals constitute the vast majority of the expelled material and the determination of the ablation parameters accessible via this method, namely the velocity distribution and the translational temperature of the particles, allow for a quantitative description of the ablation process. With this it could be shown that the ablation of the employed matrices exhibits a good reproducibility and these substances are therefore well suited for investigations using this method. Furthermore, it was discovered that Ionic Liquids containing the often used matrices 2,5-DHB and CHCA display a strong similarity in the respective ablation parameters, which are also well below their values obtained in the crystalline state. Additionally, an impact of the absorption of the laser light at the used wavelength and the energy of the irradiated photons on these parameters was found. These experiments also pointed towards the occurence of a second ablation event expelling neutral molecules with a time delay of a few hundred nanoseconds to the initial ablation. A characterization of this process and the eludication of its origin was not possible by the employment of the postionization method. To achive this, a new experiment was designed and set up, combining fast flash imaging, in which shadowgraphy pictures of the ablated particle cloud are obtained at varying times after the ablation laser pulse via the illumination by short lightpulses in the nanosecond range, and time-of-flight mass spectrometry. Based on the measurements carried out on this instrument, a model of the ablation was postulated, which explaines the delayed ablation event by the release of a part of the absorbed energy after the initial ablation is finished. The observed changes of the ablation process arising from a change in the laser fluence or the ablation wavelength can be explained by this model in a satisfactory manner.
